In a nutshell, here are the differences.

Frame texture
Backstraps
Magazine release
Recoil spring assembly

There are a couple of subtle internal differences as well.

I got midget (short) thumbs so the bigger mag release helps out a lot.

You and me both, it's nice not having to turn my whole hand just to drop the mag.

The backstraps really help me out on the 9mm frame size - I have long hands and fingers. The Gen 3 don't feel as comfortable to me.

If you are going to try to get the best trigger, still easier with the G3s. If that does not matter to you, G4 has the benefits others mentioned above and is the way to go IMHO.

You could get a Gen4 and drop a Gen3 trigger and bar in it.

What's the difference between the trigger mechanisms/bars? I've noticed my G22.3 has a better trigger pull than my G20.4, but I just chalked it up to amount of use and normal manufacturing variances.

The connector sits in the housing at a slightly different angle and that makes the pull heavier. Even if you put G3 parts in, still heavier.

My favorite Glock trigger is the fourth generation "dot" connector in a gen 3. The angle where the trigger bar moves against the connector is kind of between the standard connector and the "minus" connector.

Apart from the texture difference, ambidextrous magazine release, and dual recoil spring giving slightly less recoil, it's the tight lockup and accuracy for the Gen 4. The lockup is way tighter in the Gen 4's and the accuracy is seen extremely fast at extended ranges for a handgun. They don't come close to me. The Gen 4 is way above all previous generations on the accuracy alone.
