No, go ahead
and carry. With a permit the law doesn't apply to you on public transportation. RTD posted the signs as a reiteration of state law, that it is a SPECIFIC crime to carry aboard public transportation - sorta makes people feel safe and fuzzy/warm.
The only places that can ban it are PRIVATE property owners and/or fixed buildings that have a screening process/metal detector that is staffed.
RTD is not a private entity. Costco can (and does) prohibit concealed carry, for instance.
I hope she carries a taser too, I'd hate to think how hard a sight picture would get on a moving bus! I'd also suggest finding an old IIa front vest panel to sew into the interior of whatever bag she is carrying. They're pretty lightweight and offer good protection at need.
There was discussion about this back when CCW became "shall issue" and the position of RTD was then, and is now, that a lawful permit carrier can ride armed. Is there a specific statement allowing it? No, they think if they dont post about it, then they cant be sued for it.

I like the idea of sewing a vest into a brief case. I should tear out the pad on my laptop case and replace it with vest parts!

I parked in a RTD garage today and there's a sign (I wish I took a picture)
it says that by entering, you agree to be subject to a vehicle search
blah, blah, blah.
No ILLEGAL weapons- assuming that your wife has a carry permit (or in theory is open carrying) she is not illegal so it's not an issue

If you read the law, its says you can't carry a loaded firearm. It say nothing about carrying an unloaded firearm. :) Just keep a loaded mag with you :)

Ok, I checked at work, and this is the answer I received:
It is allowed publicly, unless posted.
My return answer was, it's posted on the bus, no weapons and received this answer back:But frankly, think about it – if you don’t have to use it, no problem. If you DO have to use it, they would likely be grateful that you did. I’m just saying.
So, take that how you may. :)
I'll present the legal garbage to this same person and see what comes of it and let you know.

Out of sight, out of mind.

I'm confused... I was under the impression that carrying a weapon was mandatory on RTD, and if you did not have one, they would issue one for the duration of your ride...
"Please be careful of the step down, place any loaner weapons in the bin, and have a nice day...."

Update: Based on the legal:
18-9-118. Firearms, explosives, or incendiary devices in facilities of public transportation.
A person commits a class 6 felony if, without legal authority, he has any loaded firearm or explosive or incendiary device, as defined in section 9-7-103, C.R.S., in his possession in, or carries, brings, or causes to be carried or brought any of such items into, any facility of public transportation, as defined in section 18-9-115 (4).
It is allowed with permit.
(i'm no lawyer though)

Has anyone or does anyone know anyone with a CCW who has been cited for carrying on RTD?
Past performance is not always an indicator of the future, however, if it hasn't happened before, it isn't likely that it is on RTD's radar.
Anyone have any regular contact with RTD security? I have spoken with them occasionally. Next time I talk with them I will ask the question. No idea when that will happen.
